Although astrocytic Pgp expression is generally undetectable in normal rat brain, up-regulation follows intracerebroventricular injection with neurotoxic concentrations of kainate (Zhang et a1 1999). Taken together, these studies suggest that activation of brain endotheliurn and astrocytes by damage, isolation or stress can alter the transporter phenotype, leading to a condition in which the primary endothelial barrier is supported by a ‘second line of defence’, the astrocytes. There are clearly implications for the epileptic brain and for treatment with AEDs.
